Each chapter is a comprehensive and well-referenced review focusing on recent developments in site-specific drug therapy, using mainly absorbable polymers. Fundamental concepts, mathematical modeling, drug-delivery technologies, preclinical studies, and clinical data where available, are covered in virtually all chapters. Extensive reference lists conclude each chapter, incorporating a few 1993 publications. A few lapses in proofing or editing e.g., on p. 45, reference 64 is missing ; are not sufficient to be distracting. The organization from historical to contemporary ; and writing style of each chapter are excellent. Sadly, we are constantly reminded throughout the book that what works in the laboratory all too often fails to work in the clinic. Yet, many new developments in drug delivery systems currently under investigation hold considerable promise of being clinically useful. The first three chapters cover fundamental concepts of chemistry, biocompatibility, tissue-drug distribution, and dosage forms of absorbable polymeric drug implants. The last chapter of the book deals with regulatory affairs governing the testing and introduction of new delivery systems. Of the specialized topics, four chapters deal with drug delivery to the brain and peripheral nerves. Although the focus is on drug delivery to tumor sites, potential treatments of other neurological diseases are considered along with novel delivery systems for local anesthetics. A chapter on drug delivery to the eye is particularly fascinating. A chapter focusing primarily on hepatocellular carcinoma, describes the structural defects in blood vessels feeding the tumor which makes them more permeable to polymer-conjugated and solubilized polymer-conjugated anticancer drugs. Perivascular drugdelivery systems for restenosis is covered in another chapter, and yet another deals with cardiovascular drug delivery systems including surfactant derivatization, polymer implants, and iontophoresis. Targeting of drugs to the lungs and pleural cavity via airways and via the pulmonary circulation are covered in a chapter devoted to pulmonary drug delivery. Two extensive chapters on drug targeting to bone marrow are especially revealing. Passive to circumvent liver influence ; and active direct targeting to bone marrow cell receptors ; drug delivery are discussed with respect to delivery of antibiotics and immunomodulators to the bone marrow. Drug delivery to the gastrointestinal tract, and prevention of surgical adhesions by use of barriers as well as by local and controlled delivery of drugs, are covered in separate chapters. This book is highly recommended for industrial and academic researchers in the field of drug-delivery technologies.
